In der Human- und Tiermedizin werden Antibiotika als Arzneimittel zur Behandlung bakterieller Infektionskrankheiten eingesetzt.<\/p>\n\n\n\n<h2 class=\"wp-block-heading\">Entstehung von Antibiotikar\u00fcckst\u00e4nden und Gefahren f\u00fcr den Verbraucher<\/h2>\n\n\n\n<p>Der Einsatz von Antibiotika als Tierarzneimittel zur Behandlung erkrankter Tiere ist unvermeidbar. Als Folge von Anwendungsfehlern, wie z.B. Nichteinhaltung der gesetzlich vorgeschriebenen Wartezeit oder bei zumeist illegalem Einsatz als Masthilfsmittel bei Tieren die der Nahrungsmittelproduktion dienen, k\u00f6nnen Antibiotikar\u00fcckst\u00e4nde in Lebensmittel tierischen Ursprungs wie z.B. Fleisch, Milch oder Eiern gelangen.<\/p>\n\n\n\n<p>Aufgrund der potentiell kanzerogenen oder toxischen Eigenschaften von Antibiotikar\u00fcckst\u00e4nden bzw. deren allergischen Potentials, stellt der Verzehr belasteter Lebensmittel f\u00fcr den Verbraucher ein direktes Gesundheitsrisiko dar. Au\u00dferdem steht der Einsatz von Antibiotika im Verdacht die Multiresistenz von pathogenen Erregern gegen in der Humanmedizin eingesetzter Antibiotika zu f\u00f6rdern.<\/p>\n\n\n\n<h2 class=\"wp-block-heading\">Gesetzliche Regelungen<\/h2>\n\n\n\n<p>Aus Gr\u00fcnden des Verbraucherschutzes wurden deshalb in vielen L\u00e4ndern R\u00fcckstandsh\u00f6chstmengen (engl.: Maximum Residue Limits, MRLs) f\u00fcr pharmakologisch wirksame Stoffe festgelegt, deren Einhaltung in unterschiedlichen \u00dcberwachungsprogrammen kontrolliert wird (Richtlinie 96\/23\/EG; Verordnung (EG) 470\/2009 und 37\/2010; Entscheidung 2003\/181\/EG). Mit dem neuen kompetitiven RIDASCREEN\u00ae Bacitracin-Test zur quantitativen Bestimmung von Futtermitteln und Urin k\u00f6nnen somit R\u00fcckst\u00e4nde dieses Antibiotikums dokumentiert werden. Die Standardreihe umfasst die Grenzwerte der EU sowie der Russischen Zollunion f\u00fcr die R\u00fcckstandsh\u00f6chstmengen von Bacitracin in Lebensmitteln. Die Nachweisgrenzen liegen unter den gesetzlichen Vorgaben. Der Test erlaubt deshalb eine sensitive sowie hochspezifische als auch einfache und schnelle R\u00fcckstandsbestimmung.<\/p>\n<\/div><div class=\"block-tabs__content  tab--2\"><p>Chinolone (oder Quinolone) sind eine Gruppe synthetischer Breitspektrum-Antibiotika. Sie unterdr\u00fccken das Gyrase-Enzym und sind daher auch als Gyrasehemmer bekannt.<\/p>\n<p>Gyrasehemmer werden in 4 Untergruppen unterteilt. Ein Gro\u00dfteil der Chinolone geh\u00f6rt zur Untergruppe der Fluorchinolone, bei denen sich eine Fluorgruppe am zentralen Ring an der Position 6 befindet. Diese Fluorchinolone geh\u00f6ren zur sogenannten zweiten Generation von Chinolonen. Einige dieser Antibiotika sind in der Veterin\u00e4rmedizin bei lebensmittelliefernden Tieren zugelassen. Fluorchinolone weisen eine sehr hohe Wirksamkeit gegen eine Vielzahl von bakteriellen Erregern auf und werden in der Veterin\u00e4rmedizin besonders h\u00e4ufig bei Rindern, Schweinen und H\u00fchnern angewendet. Der Einsatz von Fluorchinolonen hat in den vergangenen Jahren zugenommen, da die Chinolone in gro\u00dfen Mengen eingesetzt wurden, um auch Infektionskrankheiten, besonders in der H\u00fchner-, Schweine- und Fisch- bzw. Shrimpszucht, zu verhindern. Der umfangreiche Einsatz hat zu Resistenzbildungen gef\u00fchrt. Deshalb wurden in der Europ\u00e4ischen Union f\u00fcr einige Stoffe dieser Gruppe MRLs (Maximum Residue Limits) im Rahmen der Anpassung der Verordnung (EEC) No 2377\/90 eingef\u00fchrt.<\/p>\n<\/div><div class=\"block-tabs__content  tab--3\"><p>Chloramphenicol ist ein Breitbandantibiotikum, das aufgrund seiner ausgezeichneten antibakteriellen und pharmakokinetischen Eigenschaften h\u00e4ufig in der Tierzucht eingesetzt wird. Beim Menschen f\u00fchrt es jedoch zu unerw\u00fcnschten Nebenwirkungen. Aus diesem Grund wurde Chloramphenicol f\u00fcr die Behandlung von zur Lebensmittelgewinnung genutzten Tiere verboten.<\/p>\n<p>Chloramphenicol (2,2-dichloro-N-[(1R,2R)-1,3-dihydroxy-1-(4-nitrophenyl)propan-2-yl]acetamide) wird von Streptomyces venezuelae produziert. Es wurde 1947 entdeckt und wird seit den 1950er Jahren in der Veterin\u00e4rmedizin eingesetzt. Heute ist der Einsatz von Chloramphenicol (CAP) bei Tieren, die zur Lebensmittelgewinnung genutzt werden, in der EU und vielen anderen L\u00e4ndern verboten. Dennoch wird es aufgrund seiner guten Wirkung und des g\u00fcnstigen Preises h\u00e4ufig in der Viehzucht angewendet. Das Wirkprinzip von Chloramphenicol ist bakteriostatisch, wodurch es die Proteinsynthese in bakteriellen Ribosomen hemmt. Beim Menschen k\u00f6nnen Chloramphenicol-R\u00fcckst\u00e4nde eine Myelosuppression (Sch\u00e4digung des Knochenmarks) sowie eine potenziell t\u00f6dliche Chloramphenicol-induzierte aplastische An\u00e4mie verursachen. Es konnte keine unbedenkliche R\u00fcckstandsh\u00f6chstmenge festgesetzt werden. Um Verbraucher zu sch\u00fctzen, gilt daher f\u00fcr Chloramphenicol-R\u00fcckst\u00e4nde eine Null-Toleranz. F\u00fcr Testsysteme hat die EU eine Mindestleistungsgrenze (Minimum Required Performance Limit, MRPL) von 0,3 ppb festgelegt.<\/p>\n<p>Die chemische Struktur von Chloramphenicol hat 8 verschiedene Stereoisomere, von denen nur einer (RR-p-CAP) biologisch aktiv ist. Unser Test-Kit RIDASCREEN\u00ae Chloramphenicol ist spezifisch f\u00fcr dieses Stereoisomer. Der quantitative ELISA-Test ist validiert f\u00fcr eine Vielzahl von Matrizes (Milch, Milchpulver, Milchprodukte, Honig, Fleisch, Fisch, Garnelen, Eier, Futtermittel, Plasma und Serum). Die neue Version des Tests (Art. Nr. R1511) kann zudem den Metabolit Chloramphenicol glucuronide nachweisen, was das direkte Screening von Urin erm\u00f6glicht.<\/p>\n<\/div><div class=\"block-tabs__content  tab--4\"><p>Nitrofurane sind synthetische Breitbandantibiotika, die aufgrund ihrer hervorragenden antibakteriellen und pharmakokinetischen Eigenschaften h\u00e4ufig in der Tierproduktion eingesetzt werden. Sie werden aber auch zur Wachstumsbeschleunigung in der Shrimps-, Gefl\u00fcgel- und Schweineproduktion eingesetzt.<\/p>\n<p>In Langzeitversuchen mit Labortieren wurden bei der Muttersubstanz und deren Metaboliten karzinogene und mutagene Eigenschaften beobachtet. Dies f\u00fchrte zu einem Anwendungsverbot von Nitrofuranen bei Tieren, die der Lebensmittelgewinnung dienen. Die Behandlung von Nutztieren mit den Nitrofuran-Antibiotika Furaltadon, Nitrofurantoin und Nitrofurazon wurde 1993 in der EU verboten, das Verbot von Furazolidon folgte 1995. Die R\u00fcckstandsanalytik von Nitrofuranen basiert auf der Detektion der gewebegebundenen Metabolite der Muttersubstanzen. Da die Muttersubstanzen sehr schnell metabolisiert werden, sind sie nach kurzer Zeit nicht mehr nachweisbar. Die Nitrofuran-Metabolite sind jedoch noch lange nach Verabreichung nachweisbar und werden deshalb zur R\u00fcckstandskontrolle von Nitrofuranen bestimmt.<\/p>\n<p>Es gibt verschiedene Arten von Nitrofuranen:<\/p>\n<ul>\n<li>Nitrofurantoin: 1-Aminohydantoin (AHD)<\/li>\n<li>Furaltadon: 3-Amino-5-Morpholinomethyl-2-Oxazolidinon (AMOZ)<\/li>\n<li>Furazolidon: 3-Amino-2-Oxazolidinon (AOZ)<\/li>\n<li>Nitrofurazon: Semicarbazid (SEM)<\/li>\n<\/ul>\n<p>Vor der Analyse m\u00fcssen die Metabolite mittels Inkubation mit 2- Nitrobenzaldehyd in NP-AHD, NP-AMOZ, NP-AOZ und NP-SEM derivatisiert werden.<\/p>\n<\/div><div class=\"block-tabs__content  tab--5\"><p>\u03b2-Lactam-Antibiotika welche nach Ihrem charakteristischen f\u00fcnfgliedrigen \u03b2\u2011Lactam-Ring benannt sind, stellen eine gro\u00dfe Gruppe von Antibiotika dar. Sie werden anhand ihrer chemischen Struktur in mehrere Untergruppen klassifiziert, von denen die Penicilline, Cephalosporine und Carbapeneme die wichtigsten sind.<\/p>\n<p>Die Penicilline wurden im Jahre 1928 von Alexander Fleming eher versehentlich entdeckt und hemmen die Zellwandsynthese von grampositiven Bakterien. Trotz ihrer fr\u00fchen Entdeckung, sind sie weltweit immer noch die am h\u00e4ufigsten eingesetzte Gruppe von Antibiotika. Speziell f\u00fcr Mastitis, eine bakterielle Infektion der Euter von Milchk\u00fchen, werden sie zur Prim\u00e4rbehandlung eingesetzt. Aus Gr\u00fcnden des Verbraucherschutzes und zur Absicherung der Prozesssicherheit in der Milchindustrie, wurden in der Verordnung (EU) Nr. 37\/2000 R\u00fcckstandsh\u00f6chstmengen (MRLs) f\u00fcr 7 Penicilline in Lebensmitteln tierischen Ursprungs festgelegt.<\/p>\n<\/div><div class=\"block-tabs__content  tab--6\"><p>Streptomycin geh\u00f6rt neben Penicillin zu den in der Veterin\u00e4rmedizin im Rahmen der Mastitis-Therapie am h\u00e4ufigsten eingesetzten Antibiotika. Bei Nichteinhaltung der gesetzlich vorgeschriebenen Wartezeit sowie bei unsachgem\u00e4\u00dfer Anwendung k\u00f6nnen Streptomycin-R\u00fcckst\u00e4nde in Lebensmitteln tierischen Ursprungs auftreten.<\/p>\n<p>In hohen Konzentrationen wirkt Streptomycin ototoxisch und nephrotoxisch. Im Zusammenhang mit Lebensmitteln sind jedoch die chronische Belastung mit niedrigen Konzentrationen und die damit verbundenen allergischen Reaktionen, die m\u00f6gliche Sch\u00e4digung der Darmflora sowie die Induzierung von Resistenzen bei pathogenen Mikroorganismen von gr\u00f6\u00dferer Bedeutung. Um gesundheitliche Risiken f\u00fcr den Konsumenten zu vermeiden und lebensmitteltechnologische Probleme zu verhindern, ist somit ein empfindliches und einfaches Verfahren zum Nachweis von Streptomycin n\u00f6tig. In der EU-Verordnung sind die H\u00f6chstwerte f\u00fcr Streptomycin unter den Tierarzneimittelr\u00fcckst\u00e4nden in Fleisch und Milch festgelegt: Muskel und Leber: 500 ppb, Nieren: 1000 ppb, Honig: 10 ppb und Milch: 200 ppb.<\/p>\n<\/div><div class=\"block-tabs__content  tab--7\"><p>Sulfonamide werden in der K\u00e4lber- und Schweinemast h\u00e4ufig als Futtermittelzusatzstoffe eingesetzt. Zusammen mit Antifolaten (Dihydrofolat-Reduktase-Hemmer) wie z. B. Trimethoprim und Pyrimethamin werden sie in der Veterin\u00e4rmedizin auch h\u00e4ufig zur Behandlung von Darminfektionen, Mastiden, Lungenentz\u00fcndungen und anderen systemischen Infektionen verwendet. Die Kontamination von Lebensmitteln tierischen Ursprungs, wie Milch und Fleisch, mit Sulfonamiden ist deshalb m\u00f6glich und wurde auch h\u00e4ufig (vor allem im Ausland) nachgewiesen. Laut EU-Recht gilt die H\u00f6chstmenge f\u00fcr alle Stoffe der Sulfonamidgruppe von 100 \u00b5g\/kg in Muskel, Fett, Leber und Niere sowie von 100 \u00b5g\/l in Milch.<\/p>\n<\/div><div class=\"block-tabs__content  tab--8\"><p>Aureomycin (Chlortetracyclin) wurde 1948 von Duggan als Stoffwechselprodukt der Aktinomycetenspezies Streptomyces aureofaciens isoliert. Dies war das erste Antibiotikum aus der Gruppe der Tetracycline.<\/p>\n<p>In Deutschland sind Tetracyclin, Chlortetracyclin und Oxytetracyclin in der Veterin\u00e4rmedizin zugelassen. Deshalb k\u00f6nnen Gesundheitsgef\u00e4hrdungen f\u00fcr den Verbraucher entstehen. Nach EU-Recht, Verordnung Nr. 37\/2010 sind f\u00fcr Tetracyclin (die Summe von Muttersubstanz und ihrem 4-Epimer) in allen zur Lebensmittelerzeugung genutzten Tierarten folgende R\u00fcckstandsh\u00f6chstmengen festgelegt: 100 ppb im Muskel und in Milch.
