L\u2019utilizzo non corretto di antibiotici nell\u2019allevamento e nella produzione di cibo, inoltre, aumenta indirettamente la resistenza dei batteri patogeni ai vari antibiotici utilizzati per la cura delle afflizioni umane. Prende il nome dal ceppo di Bacillus che la produce e da \u201cTracy\u201d, una ragazzina di 7 anni; la bacitracina \u00e8 stata infatti isolata per la prima volta in una sua frattura tibiale esposta, nel 1945.<\/p>\n<p>La bacitracina \u00e8 composta da molti polipeptidi differenti, il pi\u00f9 biologicamente attivo dei quali \u00e8 la bacitracina A. Vanta infatti, grazie alle sue caratteristiche di inibitore della sintesi della parete cellulare, effetti battericidi contro un gran numero di batteri gram-positivi e gram-negativi. Oltre alle sue applicazioni in campo veterinario, la bacitracina viene utilizzata come promotore delle crescita antimicrobico nei processi di allevamento animale. I suoi residui possono quindi essere presenti in molti alimenti di origine animale e porre un rischio concreto per la salute dei consumatori. Un utilizzo improprio degli antibiotici promuove inoltre la formazione di batteri resistenti agli antibiotici, a loro volta considerati un rischio per la salute pubblica. La bacitracina \u00e8 stata quindi eliminata, tramite il regolamento 2821\/98\/EC, dall\u2019elenco degli additivi per mangimi approvati (Direttiva 70\/524\/EEG, Appendice B), e sono stati imposti limiti massimi alla presenza di suoi residui negli alimenti (Regolamento 37\/2010\/EC).<\/p>\n<\/div><div class=\"block-tabs__content  tab-residuestabs1-2\"><p>I chinoloni sono una famiglia di farmaci antibiotici sintetici ad ampio spettro, in grado di inibire l\u2019enzima DNA girasi e noti quindi anche con il nome di inibitori della DNA girasi.<\/p>\n<p>Gli inibitori della DNA girasi si suddividono in quattro gruppi. La maggior parte appartiene al gruppo dei fluorochinoloni, che si distinguono per la presenza un atomo di fluoro sull\u2019anello centrale, solitamente in posizione 6; sono anche detti chinoloni di seconda generazione. Alcuni trovano applicazione come antibiotici nel campo della medicina veterinaria per gli animali da produzione alimentare. I fluorochinoloni sono in grado di contrastare un ampio spettro di specie batteriche. Sono molto utilizzati nella cura di animali da allevamento come bovini, suini e pollame. L\u2019utilizzo dei fluorochinoloni \u00e8 aumentato negli ultimi anni per sopperire alla crescente necessit\u00e0 di prevenzione di malattie infettive, in particolare nell\u2019allevamento di pollame, suini, pesci e gamberi. Questo ampio utilizzo ha portato a un aumento della resistenza microbica agli antibiotici, e di conseguenza all\u2019emendamento del Regolamento del Consiglio (EEC) 2377\/90 e all\u2019introduzione di LMR (limiti massimi di residui) per alcuni fluorochinoloni.<\/p>\n<\/div><div class=\"block-tabs__content  tab-residuestabs1-3\"><p>Il cloramfenicolo \u00e8 un antibiotico ad ampio spettro molto utilizzato nell\u2019industria zootecnica per le sue eccellenti propriet\u00e0 antibatteriche e farmacocinetiche. Negli esseri umani pu\u00f2 causare effetti ematotossici indesiderati, in particolare l\u2019anemia aplastica indotta da cloramfenicolo, per la quale non \u00e8 ancora stata stabilita una precisa relazione dose-effetto. Ne \u00e8 stato quindi proibito l\u2019utilizzo nel trattamento di animali utilizzati nella produzione alimentare.<\/p>\n<p>Il cloramfenicolo (2,2-dicloro-N-[(1R,2R)-1,3-diidrossi-1-(4-nitrofenil)propan-2-il]acetammide), prodotto da Streptomyces venezuelae, \u00e8 stato scoperto nel 1947 e utilizzato in medicina veterinaria fin dagli anni \u201950. Oggi, nell\u2019UE e in altri Paesi l\u2019uso dell\u2019antibiotico ad ampio spettro cloramphenicolo (CAP) negli animali da produzione alimentare \u00e8 proibito. Il CAP trova comunque ampio utilizzo nell\u2019industria alimentare per le eccellenti propriet\u00e0 antibatteriche e farmacocinetiche e per il prezzo ridotto. Il cloramfenicolo ha effetti batteriostatici in quanto inibitore della sintesi delle proteine dei ribosomi batterici. Negli esseri umani i residui di CAP possono causare mielosoppressione (soppressione del midollo osseo) e anemia aplastica, una malattia potenzialmente mortale. Non \u00e8 stato possibile stabilire il livello di residuo minimo per evitare questi effetti collaterali. Per tutelare la salute dei consumatori, quindi, \u00e8 stato vietato l\u2019utilizzo del cloramfenicolo negli animali utilizzati nella produzione alimentare e implementata una politica di tolleranza zero nei confronti dei residui di cloramfenicolo. L\u2019UE impone un limite minimo di rendimento richiesto (LMRR) pari a 0,3 \u03bcg\/kg per tutti i sistemi analitici.<\/p>\n<p>La struttura chimica del cloramfenicolo presenta 8 diversi stereoisomeri, uno solo dei quali (RR-p-CAP) biologicamente attivo. Il nostro test \u00e8 specifico per questo stereoisomero. Il test ELISA quantitativo RIDASCREEN\u00ae Chloramphenicol \u00e8 certificato per un\u2019ampia gamma di matrici (latte, latte in polvere, prodotti a base di latte, miele, carne, pesce, gamberi, uova, mangimi, plasma e siero). La nuova versione R1511 \u00e8 inoltre in grado di rilevare il metabolita cloramfenicolo glucuronide e permette quindi l\u2019analisi diretta delle urine.<\/p>\n<\/div><div class=\"block-tabs__content  tab-residuestabs1-4\"><p>I nitrofurani sono antibiotici sintetici ad ampio spettro molto utilizzati nell\u2019industria zootecnica per le loro eccellenti propriet\u00e0 antibatteriche e farmacocinetiche. Trovano inoltre utilizzo come promotori della crescita nell\u2019allevamento di gamberi, pollame e suini.<\/p>\n<p>Esperimenti a lungo termine sugli animali hanno dimostrato il potenziale cancerogeno e mutageno dei composti progenitori e dei loro metaboliti, portando al divieto di utilizzo di nitrofurani nel trattamento di animali da produzione alimentare. Nel 1993 l\u2019UE ha vietato l\u2019utilizzo dei nitrodurani furaltadone, nitrofurantoina e nitrofurazone, ai quali si \u00e8 aggiunto nel 1995 il furazolidone. L\u2019analisi dei nitrofurani si basa sul rilevamento dei metaboliti dei composti nitrofuranici che si fissano sui tessuti; i composti progenitori vengono infatti metabolizzati troppo rapidamente e risultano quindi difficili da rintracciare a breve distanza dal trattamento. I metaboliti, invece, sopravvivono molto pi\u00f9 a lungo dopo la somministrazione e vengono quindi utilizzati per rilevare eventuali abusi.<\/p>\n<p>Esistono diversi tipi di nitrofurani<\/p>\n<ul>\n<li><strong>Nitrofurantoina:<\/strong> 1-amminoidantoina (AHD)<\/li>\n<li><strong>Furaltadone:<\/strong> 3-ammino-5-morfolinometil-2-ossazolidinone (AMOZ)<\/li>\n<li><strong>Furazolidone:<\/strong> 3-ammino-2-ossazolidinone (AOZ)<\/li>\n<li><strong>Nitrofurazone:<\/strong> semicarbazide (SEM)<\/li>\n<\/ul>\n<p>In preparazione all\u2019analisi, i metaboliti devono essere derivatizzati mediante incubazione con 2-nitrobenzaldeide in NP-AHD, NP-AMOZ, NP-AOZ e NP-SEM.<\/p>\n<\/div><div class=\"block-tabs__content  tab-residuestabs1-5\"><p>Il vasto gruppo degli antibiotici beta-lattamici prende il nome dai caratteristici anelli a quattro atomi detti beta-lattami. Vengono suddivisi in gruppi in base alla propria struttura chimica; i pi\u00f9 importanti sono le penicilline, le cefalosporine e i carbapenemi.<\/p>\n<p>Le penicilline, scoperte accidentalmente da Alexander Fleming nel 1928, sono in grado di inibire la sintesi delle pareti cellulari dei batteri gram-positivi. Rimangono tuttora tra gli antibiotici pi\u00f9 utilizzati al mondo, per esempio per il trattamento della mastite, un\u2019infezione batterica che colpisce mammelle delle vacche da latte. Per ragioni di tutela dei consumatori e sicurezza dei processi dell\u2019industria lattiero-casearia, il Regolamento della Commissione (UE) 37\/2010 ha stabilito limiti massimi per i residui (LMR) di 7 tipi di penicilline negli alimenti di origine animale.<\/p>\n<\/div><div class=\"block-tabs__content  tab-residuestabs1-6\"><p>Insieme agli antibiotici betalattamici per uso veterinario, la streptomicina \u00e8 uno degli antibiotici pi\u00f9 utilizzati per il trattamento della mastite. \u00c8 possibile quindi trovare residui di streptomicina negli alimenti di origine animale qualora non si rispettino i tempi di ritenzione o si faccia un uso improprio dell\u2019antibiotico.<\/p>\n<p>La streptomicina mostra ad elevate concentrazioni effetti ototossici e nefrotossici. Alle basse concentrazioni, come quelle che si ritrovano negli alimenti, pu\u00f2 causare allergie, indebolire la flora intestinale e favorire lo sviluppo di microorganismi resistenti o patogeni. Per proteggere il consumatore da rischi per la salute ed evitare problemi tecnologici \u00e8 necessaria l\u2019adozione di un metodo semplice e sensibile per la rilevazione della streptomicina. Le normative dell\u2019Unione Europea per la streptomicina stabiliscono i LMS (limite massimo del residuo) per la carne e il latte (tessuto muscolare ed epatico: 500 \u03bcg\/kg, renale: 1000 \u03bcg\/kg e, per il latte, 200 \u03bcg\/l).<\/p>\n<\/div><div class=\"block-tabs__content  tab-residuestabs1-7\"><p>I sulfamidici sono largamente impiegati come additivi alimentari, principalmente per l\u2019ingrassamento di vitelli e suini. In combinazione con gli inibitori della diidrofolato reduttasi (trimetoprim, tetrossoprim, pirimetamina), i sulfamidici trovano impiego anche nella pratica veterinaria per il trattamento di infezioni intestinali, mastiti, polmoniti e altre malattie sistemiche. Pertanto \u00e8 possibile reperire residui di sulfamidici in alimenti di origine animale come le carni e il latte, specialmente se provenienti da Paesi esteri. La legislazione UE stabilisce un limite massimo di residui per tutte le sostanze appartenenti al gruppo dei sulfamidici pari a 100 \u03bcg\/kg per campioni di tessuto muscolare, adiposo, epatico e renale e di 100 \u03bcg\/l nel latte.<\/p>\n<\/div><div class=\"block-tabs__content  tab-residuestabs1-8\"><p>Nel 1948 l\u2019aureomicina (clortetraciclina) fu isolata da Duggan come metabolita di una specie di actinomiceti, <em>Streptomyces aureofaciens<\/em>. Essa fu la prima sostanza antibiotica isolata facente parte del gruppo delle tetracicline.<\/p>\n<p>In Germania la tetraciclina, la clortetraciclina e l\u2019ossitetraciclina sono autorizzate nella pratica veterinaria. Pertanto il loro uso pu\u00f2 comportare rischi per la salute del consumatore. La presenza di residui di tetracicline (somma del composto madre e dei 4 epimeri) negli animali utilizzati per l\u2019alimentazione umana \u00e8 governata, nell\u2019ambito della Comunit\u00e0 Europea, dal Regolamento No.37\/2010, che stabilisce limiti di 100 \u03bcg\/kg nel muscolo e nel latte.
