In de humane en veterinaire geneeskunde worden antibiotica daarom gebruikt als geneesmiddel voor de behandeling van bacteri\u00eble aandoeningen.<\/p>\n\n\n\n<h3 class=\"wp-block-heading\">Oorsprong en gezondheidsrisico\u2019s van antibiotica-residuen in voedingsmiddelen<\/h3>\n\n\n\n<p>Het gebruik van antibiotica als geneesmiddel voor de behandeling van zieke dieren is een kwestie van dierenwelzijn en daarom onvermijdelijk. Als gevolg van toepassingsfouten, zoals niet-naleving van de voorgeschreven wachttijd of misbruik van antibiotica ter bevordering van de groei, kunnen residuen van antibiotica voorkomen in voedingsmiddelen van dierlijke oorsprong, zoals vlees, melk of eieren.<\/p>\n\n\n\n<p>Door de mogelijk kankerverwekkende en toxische eigenschappen van antibiotica-residuen en door het allergische potentieel ervan vormt de consumptie van residubevattende voedingsmiddelen een direct risico voor de volksgezondheid. Bovendien bevordert het oneigenlijke gebruik van antibiotica in de veehouderij en de voedingsmiddelenproductie de resistentie van pathogene bacteri\u00ebn tegen meerdere antibiotica die in de humane geneeskunde worden gebruikt. Antibiotica-residuen vormen een risico voor de veiligheid van het productieproces en dus ook een economisch risico, omdat ze een remmend effect hebben op biotechnologische productieprocessen met micro-organismen, zoals startculturen in de zuivelindustrie.<\/p>\n\n\n\n<h3 class=\"wp-block-heading\">Wettelijke bepalingen<\/h3>\n\n\n\n<p>Met het oog op consumentenbescherming hebben veel landen maximum residulimieten (MRL) voor farmacologisch werkzame stoffen vastgesteld en wordt toegezien op de naleving ervan in controleprogramma\u2019s (richtlijn 96\/23\/EG; verordening (EG) nr. 470\/2009 en 37\/2010; besluit 2003\/181\/EG). Het middel werd vernoemd naar de producerende bacillusstam en naar Tracy, een 7-jarig meisje met een open tibiafractuur waaruit deze stam in 1945 werd ge\u00efsoleerd.<\/p>\n<p>Bacitracine is een mengsel van verschillende polypeptiden, waarvan bacitracine A als hoofdbestanddeel de grootste biologische activiteit heeft. Door remming van celwandsynthese heeft het een breedspectrum bactericide werking tegen zowel grampositieve als gramnegatieve bacteri\u00ebn. Naast veterinaire toepassingen kan bacitracine worden gebruikt als antimicrobi\u00eble groeibevorderaar in de veehouderij. Daardoor kunnen residuen in voedingsmiddelen van dierlijke oorsprong achterblijven en een gezondheidsrisico voor de consument vormen. Onjuist gebruik van antibiotica bevordert ook de vorming van antibiotica-resistente bacteri\u00ebn, die een toenemend probleem voor de volksgezondheid vormen. Bijgevolg is bacitracine bij verordening 2821\/98\/EG geschrapt van de lijst met goedgekeurde diervoeder-additieven (bijlage B van richtlijn 70\/524\/EEG) en zijn maximum residulimieten voor voedingsmiddelen vastgesteld (verordening 37\/2010\/EG).<\/p>\n<\/div><div class=\"block-tabs__content  tab-residuestabs1-2\"><p>De chinolonen zijn een familie van synthetische breedspectrumantibiotica. Ze remmen het gyrase-enzym en staan daarom ook bekend als gyraseremmers.<\/p>\n<p>Gyraseremmers worden onderverdeeld in vier subgroepen. De meerderheid van de chinolonen behoort tot de subgroep van fluorchinolonen, die een fluorgroep aan het centrale ringsysteem hebben, meestal op de 6e positie. De fluorchinolonen behoren tot de zogenaamde tweede-generatie-chinolonen. Sommige daarvan zijn toegelaten als antibioticum in de veterinaire geneeskunde voor gebruik bij voedselproducerende dieren. Fluorchinolonen zijn breedspectrumantibiotica tegen veel bacteriesoorten. Ze worden vaak gebruikt in de veterinaire geneeskunde, vooral bij runderen, varkens en kippen. Het gebruik van fluorchinolonen is de afgelopen jaren toegenomen omdat grote hoeveelheden van deze antibiotica werden toegepast om infectieziekten te voorkomen, vooral in de kippen- en varkenshouderij en de vis- en garnalenkwekerij. Het wijdverbreide gebruik bij voedselproducerende dieren heeft geleid tot microbi\u00eble resistenties. Dit leidde tot een wijziging van raadsverordening (EEG) nr. 2377\/90 en tot de invoering van maximum residulimieten (MRL) voor bepaalde fluorchinolonen.<\/p>\n<\/div><div class=\"block-tabs__content  tab-residuestabs1-3\"><p>Chlooramfenicol is een breedspectrumantibioticum dat veelvuldig wordt gebruikt in de dierlijke productie vanwege de uitstekende antibacteri\u00eble en farmacokinetische eigenschappen. Bij de mens leidt het middel echter tot hematotoxische bijwerkingen, met name door chlooramfenicol-ge\u00efnduceerde aplastische anemie waarvoor nog geen dosis-effectrelatie is vastgesteld. Dit heeft geleid tot een verbod op chlooramfenicol voor de behandeling van voedselproducerende dieren.<\/p>\n<p>Chlooramfenicol (2,2-dichloor-N-[(1R, 2R)-1,3-dihydroxy-1- (4-nitrofenyl)propaan-2-yl]acetamide), geproduceerd door Streptomyces venezuelae, werd voor het eerst ontdekt in 1947 en wordt sinds de jaren vijftig in de veterinaire geneeskunde gebruikt. Het gebruik van het breedspectrumantibioticum chlooramfenicol is tegenwoordig illegaal voor toediening aan voedselproducerende dieren in de EU en veel andere landen wereldwijd. Het wordt nog steeds vaak gebruikt in de dierlijke productie vanwege de uitstekende antibacteri\u00eble en farmacokinetische eigenschappen en de lage prijs. Het werkingsmechanisme van chlooramfenicol is bacteriostatisch door remming van de eiwitsynthese in bacteri\u00eble ribosomen. Bij mensen kunnen chlooramfenicolresiduen myelosuppressie (beschadiging van het beenmerg) veroorzaken en de potentieel dodelijke chlooramfenicol-ge\u00efnduceerde aplastische anemie. Voor deze bijwerkingen kon geen veilige residulimiet worden vastgesteld. Om de gezondheid van de consument te waarborgen werd chlooramfenicol daarom helemaal verboden voor de behandeling van voedselproducerende dieren en werd besloten tot een nultolerantie voor chlooramfenicolresiduen. De EU heeft voor alle testsystemen een MRPL (Minimum Required Performance Limit: minimaal vereiste prestatielimiet) van 0,3 ppb vastgesteld.<\/p>\n<p>De chemische structuur van chlooramfenicol heeft 8 verschillende stereo-isomeren, waarvan er slechts \u00e9\u00e9n (RR-p-CAP) biologisch actief is. De testkit is specifiek voor deze stereo-isomeer. De RIDASCREEN\u00ae Chloramphenicol kwantitatieve ELISA is gevalideerd voor een breed scala aan matrices (melk, melkpoeder en melkproducten, honing, vlees, vis, garnalen, eieren, diervoeder, plasma en serum). Bovendien kan de nieuwe versie R1511 ook de metaboliet chlooramfenicolglucuronide detecteren, waardoor directe screening van urine mogelijk is.<\/p>\n<\/div><div class=\"block-tabs__content  tab-residuestabs1-4\"><p>Nitrofuranen zijn synthetische breedspectrumantibiotica, die vanwege de uitstekende antibacteri\u00eble en farmacokinetische eigenschappen veelvuldig in de dierlijke productie worden gebruikt. Ze worden ook gebruikt als groeibevorderaar bij de productie van garnalen, pluimvee en varkens.<\/p>\n<p>Langdurige dierproeven hebben aangetoond dat de oorspronkelijke verbindingen en hun metabolieten kankerverwekkende en mutagene eigenschappen hebben. Dit heeft geleid tot een verbod op nitrofuranen voor de behandeling van voedselproducerende dieren. In 1993 verbood de EU het gebruik van de nitrofuranen furaltadon, nitrofuranto\u00efne en nitrofurazon voor gebruik bij dieren die als bron voor voedingsmiddelen gebruikt worden, en in 1995 werd het gebruik van furazolidon ook verboden. De analyse van nitrofuranen is gebaseerd op de detectie van weefselgebonden metabolieten van nitrofuranen. Het is moeilijk om de oorspronkelijke verbindingen nauwkeurig te detecteren omdat ze zeer snel gemetaboliseerd worden na de behandeling. De aan weefsel gebonden nitrofuraanmetabolieten blijven echter lang aanwezig en kunnen worden gebruikt om nitrofuraanmisbruik op te sporen.<\/p>\n<p>Er zijn verschillende soorten nitrofuranen:<\/p>\n<ul>\n<li><strong>Nitrofuranto\u00efne:<\/strong> 1-aminohydanto\u00efne (AHD)<\/li>\n<li><strong>Furaltadon:<\/strong> 3-amino-5-morfolinomethyl-2-oxazolidinon (AMOZ)<\/li>\n<li><strong>Furazolidon:<\/strong> 3-amino-2-oxazolidinon (AOZ)<\/li>\n<li><strong>Nitrofurazon:<\/strong> Semicarbazide (SEM)<\/li>\n<\/ul>\n<p>Voorafgaand aan de analyse moeten de metabolieten door incubatie met 2-nitrobenzaldehyde worden omgezet in NP-AHD, NP-AMOZ, NP-AOZ en NP-SEM.<\/p>\n<\/div><div class=\"block-tabs__content  tab-residuestabs1-5\"><p>B\u00e8talactamantibiotica zijn vernoemd naar de karakteristieke 5-ledige b\u00e8talactamring en vormen een grote groep antibiotica. Ze worden op basis van hun chemische structuur ingedeeld in verschillende subgroepen, met als belangrijkste de penicillinen, cefalosporinen en carbapenems.<\/p>\n<p>De penicillinen werden bij toeval ontdekt in 1928 door Alexander Fleming en ze remmen de celwandsynthese van grampositieve bacteri\u00ebn. Ondanks de vroege ontdekking vormen ze nog steeds de meest gebruikte groep antibiotica ter wereld. Met name voor mastitis, een bacteri\u00eble uier-infectie bij de melkkoe, zijn ze de eerstelijnsbehandeling. Om redenen van consumentenbescherming en procesveiligheid in de zuivelindustrie zijn in commissie-verordening (EU) nr. 37\/2010 maximum residulimieten (MRL) voor 7 penicillinen in voedingsmiddelen van dierlijke oorsprong vastgesteld.<\/p>\n<\/div><div class=\"block-tabs__content  tab-residuestabs1-6\"><p>Naast de b\u00e8talactamantibiotica is streptomycine in de veterinaire geneeskunde een van de meest gebruikte antibiotica voor de behandeling van mastitis. Residuen van streptomycine kunnen derhalve in voedingsmiddelen van dierlijke oorsprong voorkomen, indien de wachttijd niet wordt nageleefd of onjuist wordt toegepast.<\/p>\n<p>Bij hoge concentraties leidt streptomycine tot ototoxische en nefrotoxische effecten. De chronische blootstelling van mensen aan lage concentraties, zoals in voeding, kan allergie\u00ebn veroorzaken, de darmflora aantasten en resistentie van pathogene micro-organismen opwekken. Om de consument tegen gezondheidsrisico\u2019s te beschermen en om voedingstechnologische problemen te vermijden, is een gevoelige en eenvoudige methode voor de detectie van streptomycine noodzakelijk. De EU-richtlijnen voor streptomycine zijn gespecificeerd in maximum residulimieten (MRL) voor vlees en melk (spier en lever: 500 ppb, nier: 1000 ppb, honing: 10 ppb en melk: 200 ppb).<\/p>\n<\/div><div class=\"block-tabs__content  tab-residuestabs1-7\"><p>Sulfonamiden worden op grote schaal gebruikt als diervoeder-additieven, voornamelijk voor het vetmesten van kalveren en varkens. Gecombineerd met remmers van dihydrofolaatreductase zoals trimethoprim, tetroxoprim of pyrimethamine worden sulfonamiden ook gebruikt in de veterinaire geneeskunde voor de behandeling van darminfecties, mastitis, longontsteking en andere (systemische) ziekten. Sulfonamideresiduen kunnen derhalve voorkomen in voedingsmiddelen van dierlijke oorsprong, zoals vlees en melk. Vooral de overdracht van het kankerverwekkende sulfamethazine vormt een bedreiging voor de volksgezondheid. Volgens EU-wetgeving geldt een maximum residulimiet (MRL) voor alle stoffen van de sulfonamidegroep van 100 \u00b5g\/kg (ppb) in spier, vet, lever en nier en van 100 \u00b5g\/l (ppb) in melk.<\/p>\n<\/div><div class=\"block-tabs__content  tab-residuestabs1-8\"><p>In 1948 werd aureomycine (chloortetracycline) door Duggan ge\u00efsoleerd als metaboliet van de actinomyces-soort Streptomyces aureofaciens. Dit was het eerste antibioticum uit de groep van tetracyclinen.<\/p>\n<p>Wereldwijd zijn tetracycline, chloortetracycline en oxytetracycline toegelaten voor veterinair gebruik. Daarom kunnen er gezondheidsrisico\u2019s voor de consument ontstaan. Voor alle voedselproducerende diersoorten worden tetracyclineresiduen (de som van de moedersubstantie en de 4-epimeer) in de EU-wetgeving, commissie-verordening (EU) nr. 37\/2010 gelimiteerd met de volgende MRL\u2019s: 100 ppb in spier en melk.
