Real-time PCR
PCR (Polymerase Chain Reaction) is een belangrijke analysemethode in zowel de voedingsmiddelenanalyse alsook de medische diagnostiek.
Een real-time PCR-test dupliceert en analyseert specifieke DNA-sequenties. In de voedingsmiddelenanalyse worden PCR-testen in veel toepassingen gebruikt, bijvoorbeeld voor de detectie van pathogene micro-organismen, voor allergeenidentificatie, voor de detectie van genetisch gemodificeerde organismen (GMO: Genetically Modified Organism) of voor de identificatie van diersoorten. PCR-testen bieden vele voordelen: Ze zijn zeer specifiek, gevoelig, robuust, snel en betrouwbaar. Bovendien kunnen de testen worden geautomatiseerd.

Real-time PCR explained
De eerste stap van elke analyse bestaat uit representatieve bemonstering en homogenisatie. Na lysis van de organische cellen in voedingsweefsel wordt het DNA geëxtraheerd met behulp van verschillende specifieke DNA- of RNA-extractiekits, ofwel voor bacteriën dan wel voor virussen.
SureFood® PREP Basic en SureFood® Prep Advanced kits worden gebruikt voor vaste voeding en diervoeder afkomstig van planten of vlees. Voor dranken worden specifieke DNA-extractiekits gebruikt afhankelijk van de matrix, terwijl de nieuwe QuickGEN lijn een gemakkelijke recht-toe-recht-aan uitvoering mogelijk maakt.
Het modulaire systeem van DNA-extractie en DNA-detectie kan onafhankelijk worden gebruikt. Amplificatie van het doel-DNA vereist naast de relevante kits het gebruik van een geschikte thermocycler met twee (FAM/HEX) tot vier (FAM/HEX/ROX/Cy5) detectiekanalen.
Toepassingsgebieden voor PCR bij het testen van voeding
Voedselallergieën kunnen ernstige immunologische reacties veroorzaken. Om de consumenten te beschermen, moeten voedingsmiddelen worden getest op allergene stoffen. Wij bieden een breed assortiment aan van PCR-kits voor de detectie van allergenen, zoals soja, hazelnoot, pinda, gluten, amandel, selderij, sesam, mosterd, vis en schaaldieren. Wij bieden ook multiplex-testen aan voor de gelijktijdige detectie van meerdere allergenen (pinda/hazelnoot/walnoot en selderij/soja/mosterd).
Specificatie van soorten is van groot belang: met vleesproducten is fraude mogelijke middels onjuiste etikettering (zie het paardenvleesschandaal). Een andere toepassing is de nalevingsgarantie van religieuze voedingsregels (halal, koosjer). Onze SureFood® Animal ID productlijn maakt gevoelig testen voor producten van varken, rund, paard, ezel, kip, kalkoen en andere soorten mogelijk. Met de SureFood® Fish ID productlijn bieden we een breed assortiment aan van PCR-kits voor de identificatie van vissoorten.
Bij de productie van dranken is de detectie van drankbedervers van cruciaal belang. Met de GEN-IAL® productielijn bieden we een breed assortiment aan van screeningkits voor de detectie van bederforganismen in de bier- en wijnproductie, bijvoorbeeld Lactobacillus, Pediococcus, Megasphaera, Pectinatus, Dekkera bruxellensis en Oenococcus oeni. De testkits kunnen worden gebruikt voor zowel controle tijdens het proces, alsook de eindproductcontrole.
De afgelopen jaren is de wereldwijde productie van genetisch gemodificeerde voeding gestaag toegenomen. Omdat voedingsmiddelen die genetisch gemodificeerde organismen (GMO: Genetically Modified Organism) bevatten, moeten worden geëtiketteerd, neemt de vraag naar gevoelige testmethoden toe. Onze SureFood® GMO productlijn maakt de kwalitatieve en kwantitatieve detectie van GMO’s mogelijk – zoals genetisch gemodificeerde mais, LibertyLink-rijst of Roundup Ready-soja.
De detectie van microbiologische besmetting is een van de voornaamste toepassingsgebieden van PCR in de voedingsmiddelenanalyse. Onze productlijn SureFast® detecteert een breed scala aan pathogene bacteriën (bijv. Salmonella, Listeria, Legionella, E. coli, Staphylococcus aureus, Campylobacter), virussen (bijv. Norovirus, Hepatitis en Influenza), gisten en schimmels in voeding.
