PCR en temps réel
La PCR (abréviation de polymerase chain reaction ou réaction en chaîne par polymérase) est une méthode d’analyse importante dans le domaine de l’analyse des aliments et du diagnostic médical.
La PCR en temps réel permet de dupliquer et d’analyser des séquences d’ADN spécifiques. Dans le domaine de l’analyse des aliments, les tests de PCR sont utilisés dans de nombreux contextes : détection de micro-organismes pathogènes, identification d’allergènes, détection d’organismes génétiquement modifiés (OGM) ou identification d’espèces animales, par exemple. Les tests PCR procurent de nombreux avantages : Ils sont très spécifiques, sensibles, robustes, rapides et fiables. Ils peuvent, par ailleurs, être automatisés.

Real-time PCR explained
La première étape de toute analyse est d’effectuer un échantillonnage représentatif et d’homogénéiser les échantillons. Après lyse cellulaire des tissus alimentaires, l’ADN est préparé à l’aide de différents kits de préparation de l’ADN ou de l’ARN pour rechercher des bactéries ou des virus.
Les kits SureFood® PREP Basic et SureFood® Prep Advanced permettent de traiter des denrées et aliments solides d’origine végétale ou animale. Pour les boissons, des kits spécifiques de préparation de l’ADN sont utilisés en fonction des matrices qui peuvent être manipulées en toute simplicité grâce à la nouvelle gamme QuickGEN.
Le système modulaire de préparation de l’ADN et de détection de l’ADN peut être utilisé indépendamment. En plus des kits adéquats, l’amplification de l’ADN cible nécessite l’utilisation d’un thermocycleur approprié comportant deux (FAM/HEX) et jusqu’à 4 (FAM/HEX/ROX/Cy5) canaux de détection.
Les allergies alimentaires peuvent provoquer des réactions immunologiques graves. Afin de protéger les consommateurs, les produits alimentaires doivent être testés pour vérifier l’absence de substances allergènes. Nous proposons un large éventail de kits de PCR pour la détection de différents allergènes : soja, noisettes, arachides, gluten, amande, céleri, sésame, moutarde, poisson ou crustacés. Nous proposons également des tests multiplex pour la détection simultanée de plusieurs allergènes (arachides/noisettes/noix et céleri/soja/moutarde).
La détermination des espèces présente un grand intérêt : en effet, les produits carnés peuvent faire l’objet de déclarations frauduleuses comme lors du scandale de la viande de cheval. Le respect des règles religieuses (nourriture halal ou kascher) fait également partie des applications possibles. Notre gamme de produits SureFood® Animal ID permet d’effectuer des tests sensibles pour le porc, le bœuf, le cheval, l’âne, le poulet, la dinde et d’autres espèces. La gamme SureFood® Fish ID inclut un large éventail de kits de PCR pour l’identification des espèces de poissons.
Lors de la fabrication de boissons, la détection des facteurs d’altération est cruciale. La gamme GEN-IAL® inclut un large éventail de kits de dépistage pour le screening des micro-organismes responsables de l’altération dans les secteurs de la bière et du vin ; par exemple, Lactobacillus, Pediococcus, Megasphaera, Pectinatus, Dekkera bruxellensis ou Oenococcus oeni. Les kits de test peuvent être utilisés pour effectuer des contrôles en cours de fabrication et/ou le contrôle du produit final.
Ces dernières années, la production mondiale d’aliments génétiquement modifiés n’a cessé d’augmenter. Les exigences relatives à l’étiquetage des produits alimentaires contenant des organismes génétiquement modifiés (OGM) entraînent une demande croissante de méthodes de test sensibles. Notre gamme de produits SureFood® GMO permet la détection qualitative et quantitative d’OGM tels que le maïs génétiquement modifié, le riz Liberty Link ou le soja Roundup Ready.
Dans le domaine de l’analyse des aliments, la détection de la contamination microbiologique est l’un des principaux champs d’application de la PCR. Notre gamme de produits SureFast® permet de détecter un large éventail de bactéries pathogènes (telles que Salmonella, Listeria, Legionella, E. coli, Staphylococcus aureus, Campylobacter), de virus (comme les norovirus, l’hépatite et la grippe), de levures et de moisissures dans les denrées alimentaires.
