Intended use

The test detects the relative beef DNA amount (Bos taurus) to the total animal DNA content (amnions) in meat samples. Therefore the kit contains two PCR systems, one for detection of a beef specific gene (Beef detection gene) and one for the detection of an animal gene (Reference gene).

General Information

The real-time PCR assay can be used with established real-time PCR instruments. The technical validation of instruments was performed on Agilent Mx3005P, Applied Biosystems 7500, Bio-Rad CFX96, Roche LightCycler® 2.0, Roche LightCycler® 480 II and Qiagen Rotor-Gene Q.

Sample preparationFor DNA-preparation of meat samples the use of SureFood® PREP Basic (Art. No. S1052) is recommended.
LOD (Detection Limit)The beef specific PCR has a limit of detection of ≤ 5 DNA-copies. The assay limit of detection depends on sample matrix, processing grade, DNA-preparation and DNA-content. The limit of quantitation depends on the concentration of the sample DNA used in the analysis. For example, if 50,000 target-sequence copies of the reference gene are present, the relative quantitation limit for beef DNA is 0.04 %.
Cross Reactivity.

Cross reactivity was observed with DNA extracts from Bison sp. (100 %) and Water buffalo (Bubalis arnee) with 0.6%.
