Intended use
SureFast® EHEC/EPEC 4plex is a multiplex real-time PCR for the direct, qualitative detection and differentiation of DNA sequences of the virulence factors stx1 (subtype a-d), stx2 (subtype a-g), eae and the E. coli and Shigella spp. virulence factor ipaH.
The test contains an Internal Control DNA (ICD) as an internal control of sample preparation procedure and to determine possible PCR-inhibition.
General Information
The real-time PCR assay can be performed with commonly used real-time PCR instruments, equipped for detection of four fluorescence emissions at the channels FAM, VIC/HEX, ROX and Cy5 at the same time. The technical validation of instruments was performed on Agilent Mx3005P, BioRad CFX 96, Roche LightCycler® 480 II, Roche cobas® z 480 Analyzer, Applied Biosystems 7500 and R-Biopharm RIDA®CYCLER.

| Sample preparation | For DNA-preparation the use of SureFast® PREP Bacteria (Art. No. F1021) or Speed PREP (Art. No. F1054) is recommended. To assess the process of bacterial growth, it is suggested to compare the samples at the beginning and at the end of the culturing (bacterial growth at Cp difference > 3). For cultivation the method of the ISO 13136 is recommended. |
| LOD (Detection Limit) | The SureFast® EHEC/EPEC 4plex real-time PCR has a limit of detection of ≤ 5 DNA copies. The assay limit of detection depends on sample matrix, processing grade, DNA preparation and DNA content. |
