Intended use

The SureFast® Salmonella Species/Enteritidis/Typhimurium 4plex is a multiplex real-time PCR for the direct, qualitative detection and differentiation of Salmonella spp., Salmonella Enteritidis and Salmonella Typhimurium.

Each reaction contains an internal amplification control (IAC).

General Information

The real-time PCR assay can be performed with commonly used real-time PCR instruments, equipped for detection of four fluorescence emissions at the channels FAM, VIC/HEX, ROX and Cy5 at the same time. The technical validation of instruments was performed on Agilent Mx3005P, Agilent AriaDx, Bio-Rad CFX96, Roche LightCycler® 480 II, Roche cobas® z 480 Analyzer, Applied Biosystems 7500, Qiagen Rotor-Gene Q and Bio Molecular Systems MIC.

Sample preparationFor DNA-preparation the use of SureFast® PREP Bacteria or SureFast® Speed PREP is recommended.

To assess the process of bacterial growth, it is recommended to compare the samples at the beginning and at the end of the culturing (bacterial growth at Cp difference >3).
LOD (Detection Limit)The SureFast® Salmonella Species/Enteritidis/Typhimurium 4plex (R&D Version) real-time PCR has a limit of detection of <= 5 DNA copies.
Cross Reactivity.

Cross reactivity was observed with DNA extracts from Salmonella Amsterdam, Salmonella Blegdam, Salmonella Kiel, Salmonella Moscow and Salmonella Nitra in the detection system for Salmonella Enteritidis.
