Intended use

The test is developed for the relative estimation of the 35S CaMV DNA amount in only Corn containing products.

Therefore the kit contains two PCR systems, one specific for the 35S CaMV promoter sequence and the other one specific for Corn (the reference gene). The detection of 35S CaMV promoter is according to German Food Law § 64 LFGB.

 

General Information

The real-time PCR assay can be used with established real-time PCR instruments. The technical verification of instruments was performed on Roche LightCycler® 480 II, Qiagen Rotor-Gene Q, Bio-Rad CFX96 Dx, Bio-Rad CFX96 Opus, R-Biopharm RIDA®CYCLER and Agilent AriaDx.

Sample preparationFor DNA-preparation of raw material the use of SureFood® PREP Basic (Art. No. S1052), SureFast® Mag PREP Food (Art. No. F1060) and for highly processed food and feed the use of SureFood® PREP Advanced (Art. No. S1053) is recommended. SureFood® PREP Add On (Art. No. S1055) is intended to be used for the extraction of DNA from raw materials as well as processed food and feed with sample weight of 2 g. It is used in conjunction with the SureFood® PREP Basic.
LOD (Detection Limit)The 35S Corn PCR has a limit of detection of ca. 5 DNA-copies. This is equivalent to approx. 0.01 % for unprocessed corn grain.
The assay limit of detection depends on sample matrix, processing grade, DNA-preparation and DNA-content.
LOQ (Limit of quantification

The limit of quantitation depends on the concentration of the sample DNA used in the analysis. For example, if 50,000 target-sequence copies of Corn specific reference gene are present, the relative quantitation limit for 35S Corn DNA is 0.1 %.
