ELISA
La technique ELISA (test d’immuno-absorption enzymatique) est une méthode à base d’anticorps. Très répandue, c’est une technologie sensible, rapide et fiable.
Avec ses kits de test ELISA, R-Biopharm s’appuie sur une méthode analytique bien établie qui répond aux exigences des laboratoires modernes. Les tests RIDASCREEN® font preuve d’une sensibilité élevée et se caractérisent par leur robustesse. Les tests RIDASCREEN®FAST permettent le criblage rapide de différents produits. Ils permettent d’analyser des échantillons de denrées alimentaires et d’aliments pour animaux en seulement 30 minutes, préparation des échantillons incluse.
Principe de la technique ELISA
Les anticorps sont des protéines produites par les cellules plasmatiques des vertébrés au sein du système immunitaire adaptatif en réponse à la présence de structures (antigènes) reconnues comme étrangères à l’organisme. Les anticorps se lient à leurs antigènes par différents moyens : interactions ioniques et hydrophobes, liaisons hydrogènes et forces de van der Waals. L’interaction entre l’anticorps et son antigène est sélective et hautement spécifique, comparable à une serrure et une clé.
Le dosage par immuno-absorption enzymatique est basé sur cette reconnaissance anticorps-antigène sélective et spécifique. Il existe de nombreux formats de tests ELISA qualitatifs ou quantitatifs. La réalisation d’un test ELISA implique au moins un anticorps spécifique pour un antigène donné. Le principe de base du test est d’immobiliser l’un de ces composants immunologiques sur une phase solide : les cavités d’une microplaque. L’analyte de l’échantillon interagit avec le système anticorps-antigène. Cette interaction peut être visualisée par les enzymes, liées à des anticorps ou antigènes secondaires, ce qui indique l’existence d’une liaison anticorps-antigène. Le substrat ajouté est converti par l’enzyme couplée, ce qui entraîne un changement de couleur mesurable à l’aide d’un spectrophotomètre.
Sandwich ELISA et ELISA compétitif
Alors que le sandwich ELISA ne détecte que les protéines intactes et les fragments de grande taille comprenant au moins deux sites de liaison, le test ELISA compétitif est capable de détecter des fragments protéiques de grande et petite taille avec un seul site de liaison. Le test ELISA compétitif peut donc être utilisé pour analyser des produits alimentaires hautement transformés.
