Micotoxinas
Micotoxinas são metabólitos tóxicos produzidos por fungos. Elas podem ter efeitos sérios agudos e crônicos na saúde de humanos e animais. A análise de micotoxinas em alimentos e rações é, portanto, necessária e frequentemente exigida por legislação. Kits de teste modernos permitem uma análise rápida qualitativa e quantitativa de micotoxinas.
Ao longo da história, as micotoxinas (como as conhecemos hoje) têm prejudicado a qualidade dos produtos agrícolas. Grandes commodities como milho, trigo, arroz, soja e amendoim podem estar contaminadas com micotoxinas, dependendo das condições de crescimento e armazenamento. Outras commodities como centeio, aveia, avelãs, figos, uvas ou especiarias também podem ser afetadas por micotoxinas. O consumo de produtos contaminados por micotoxinas pode ter efeitos agudos tóxicos na saúde de humanos e animais, assim como efeitos crônicos. Além dos custos de saúde, fatores econômicos diretos também desempenham um papel, como a diminuição da eficiência no crescimento de animais de fazenda, perdas comerciais devido a limitações de importação e exportação, e perdas diretas na receita do valor da colheita. Para proteger a saúde de consumidores e animais em todo o mundo, existem legislações para micotoxinas.

Métodos para análise de micotoxinas
A análise de micotoxinas apresenta alguns desafios únicos: as toxinas de fungos são contaminantes naturalmente presentes. Sua produção antes da colheita é afetada pelas condições climáticas, assim como pelas práticas agrícolas; após a colheita, a produção depende das condições de armazenamento. Isso coloca certos desafios na gestão de risco das micotoxinas: onde, quando, com que frequência e quantos testes devo realizar?
As micotoxinas estão distribuídas de forma muito heterogênea ao longo de um produto e podem estar presentes em níveis muito baixos. As amostras a serem testadas podem ser matrizes analiticamente simples, como trigo, milho, ou matrizes muito complexas, como especiarias, café ou misturas complexas de ingredientes presentes em rações animais. É muito importante escolher o método de amostragem e análise correto para o produto a ser testado. A R-Biopharm oferece uma ampla gama de métodos analíticos para análise de micotoxinas. ELISA (ensaios imunoenzimáticos) são precisos e fáceis de usar. Os kits de teste ELISA são a solução ideal para a medição de múltiplas amostras. A R-Biopharm oferece duas linhas de produtos para ELISA:
RIDASCREEN®FAST
Tempos curtos de incubação de apenas 8 a 15 minutos, permitindo analisar até 19 amostras em uma única determinação em uma corrida.
RIDASCREEN®
Baixo limite de detecção (LOD). Com tempos de incubação mais longos, por exemplo, 75 minutos para até 42 amostras em dupla determinação. Devido aos tempos de incubação mais longos, esses kits de teste podem ser automatizados usando, por exemplo, o ThunderBolt®.
As principais vantagens do ELISA são a rapidez para um grande número de amostras, baixo custo para configuração e mínimo treinamento necessário. O ELISA pode ser automatizado usando, por exemplo, o ThunderBolt®. A eficiência de custo para automação requer um mínimo de 4 a 5 amostras testadas por corrida de ELISA. Os resultados são medidos com um leitor de placas ELISA, como o RIDA® Absorbance 96.

Os cartões de triagem AFLACARD® e OCHRACARD® são especialmente projetados para obter resultados semi-quantitativos rápidos em commodities mais complexas.
Colunas de imunoafinidade (IAC) (por exemplo, RIDA®, EASI-EXTRACT®, PREP®, RHONE®) contêm anticorpos monoclonais, tornando-as altamente seletivas. Outras vantagens incluem:
- Excelente limpeza da amostra, especialmente em commodities complexas. Isso permite o uso de padrões baseados em solvente com detecção por LC-MS/MS.
- Eluatos limpos significam que não há necessidade de padrões correspondentes à matriz ou rotulados isotopicamente.
Colunas de extração em fase sólida (SPE) usam adsorventes sólidos selecionados para ligar componentes interferentes e pigmentos, permitindo que as micotoxinas sejam facilmente analisadas. Outros benefícios são:
- Limpeza não específica, tornando esta uma ferramenta ideal para a limpeza na análise de multi-toxinas em matrizes simples como cereais.
- Limpeza simples, rápida e econômica que oferece um excelente teste de triagem em conjunto com LC-MS/MS.
- Pode ser usado em combinação com padrões correspondentes à matriz, eliminando a necessidade de padrões isotopicamente rotulados caros.
Para alto rendimento de amostras, sistemas totalmente automatizados como o CHRONECT Symbiosis RIDA®CREST, que utilizam cartuchos de imunoafinidade, podem ser usados até 15 vezes. Com LC-MS/MS, todas as micotoxinas regulamentadas são detectadas, assim como a maioria dos metabólitos mascarados e emergentes comuns. Também é possível analisar outros parâmetros, como pesticidas e medicamentos veterinários.

Análise de múltiplas micotoxinas
As micotoxinas geralmente co-ocorrem em diferentes commodities. Não é incomum encontrar diferentes tricotecenos em um mesmo produto, às vezes até em combinação com zearalenona. Outras commodities podem conter tanto aflatoxina quanto ocratoxina ou aflatoxina e fumonisina. Métodos de análise de múltiplas micotoxinas são capazes de detectar várias micotoxinas simultaneamente. Padrões de micotoxinas
Soluções de padrões de micotoxinas são rotineiramente utilizadas na análise de micotoxinas. Elas podem ser usadas para preparar uma curva de calibração para o sistema HPLC, garantindo a determinação precisa da toxina. A solução padrão também pode ser usada para enriquecer amostras a fim de verificar a eficiência de extração da toxina de certos alimentos usando um solvente específico, ou alternativamente, para enriquecer extratos de amostras.
