Kit per la rilevazione delle contaminazioni microbiologiche
Microbiologia
Gli alimenti contaminati da microorganismi quali batteri, lieviti, muffe o virus possono costituire un rischio per il consumatore. Oltre alla rilevazione dei microorganismi patogeni è importante monitorare gli organismi inquinanti al fine di ridurre perdite durante la produzione.
I microorganismi sono creature uni- e pluricellulari di microscopiche dimensioni e pressoché onnipresenti in natura (acqua, suolo, aria, eccetera). I microorganismi possono essere naturalmente presenti negli alimenti o sulle loro superfici (per es. degli ortaggi freschi), oppure intervenire come contaminazione durante il processo di produzione dei prodotti alimentari (per esempio per carenze igieniche durante la lavorazione della carne).
Nel settore alimentare si distinguono varie diverse categorie di microorganismi:
I microorganismi patogeni devono essere del tutto assenti dai prodotti alimentari, oppure limitarsi a un numero specificato di cellule per grammo di alimento. Questi limiti differiscono in base alla matrice alimentare. (Per ulteriori informazioni – DGHM Website). In caso di superamento dei limiti si potrebbero avere gravissime conseguenze per la salute del consumatore. Pertanto è prevista una tolleranza zero per 25 g di campione alimentare per la maggior parte dei più comuni patogeni (Salmonella, Campylobacter, E. coli, eccetera).
In generale, si possono distinguere due metodi di analisi microbiologica: analisi degli alimenti e monitoraggio dell’igiene/controllo delle procedure di pulizia.
Rilevazione di microorganismi negli ambienti di produzione
I test microbiologici per il controllo delle procedure di pulizia e dello stato d’igiene rilevano, in generale, i residui alimentari presenti sulle superfici non sufficientemente pulite degli ambienti di produzione. Queste sostanze nutritive residue sono alla base della crescita di microorganismi.
La conta delle colonie negli alimenti o sulle superfici può essere eseguita utilizzando le piastre Compact Dry pronte per l’uso. Le Compact Dry sono disponibili per 13 differenti parametri di analisi. Il metodo generale su cui si basano le Compact Dry è la crescita di microorganismi su terreni di coltura standard. I test si basano su substrati cromogeni specifici che vengono convertiti in prodotti colorati dal metabolismo dei microorganismi. Oltre all’analisi di campioni alimentari diluiti, le Compact Dry sono adatte anche per tamponi di superfici o per l’incubazione di filtri a membrana da campioni liquidi.
I residui alimentari sulle superfici possono essere rilevati utilizzando test per l’analisi delle proteine (RIDA®CHECK) o tramite misurazioni dell’ATP (LuciPac™Pen). La rilevazione dei residui proteici identifica approssimativamente la fase di contaminazione mediante il cambiamento di colore del reagente di rilevazione colorimetrico. Tuttavia, la quantità relativa rispettivamente di ATP (adenosintrifosfato) o AMP (adenosinmonofosfato) può essere misurata con precisione. Una speciale reazione di bioluminescenza che richiede la presenza di ATP o AMP è registrata in un apposito luminometro (Lumitester™ PD-20). Entrambi i sistemi sono indicati per un costante controllo dell’efficacia delle procedure di pulizia. Questi controlli assicurano il mantenimento dello stato d’igiene a livello industriale. Superfici igienicamente pulite consentono di ridurre il rischio di contaminazioni del prodotto.

Rilevazione di microorganismi negli alimenti
I residui tossici dei batteri presenti in campioni di alimenti e bevande possono essere analizzati con i kit per le tossine batteriche. I formati più comuni di test per l’analisi della carica microbica negli alimenti sono i saggi ELISA, i test di real-time PCR, le piastre con terreni di coltura e le piastre con agar.
Per la rilevazione di batteri patogeni sono disponibili metodi immunoenzimatici (ELISA). I test RIDASCREEN® si basano sulla tecnologia a sandwich, in cui l’antigene (proteina di superficie batterica) viene catturato nel pozzetto di una micropiastra e rilevato aggiungendo un secondo anticorpo marcato con un enzima di rilevazione specifico. La rilevazione immunologica necessita di idonee procedure di pre-arricchimento a seconda del tipo di patogeno.
È possibile eseguire la rilevazione delle tossine batteriche direttamente da campioni alimentari (RIDASCREEN® SET Total e RIDASCREEN® SET A,B,C,D,E) oppure è possibile rilevare indirettamente batteri patogeni che producono tossine (RIDASCREEN® Verotoxin) dopo un adeguato pre-arricchimento.
La rilevazione di batteri patogeni e virus può essere eseguita tramite sistemi di biologia molecolare come i kit altamente specifici di real-time PCR della linea SureFood® PATHOGEN. Il sistema modulare consiste in una preparazione del DNA ottimizzata tramite colonne a filtro spin e la diffusissima tecnologia a sonda di idrolisi TaqMan collegata a coloranti FAM per la rilevazione. Tali kit possono essere utilizzati con tutti i tipi di termociclatore per real-time PCR comunemente disponibili. Il controllo interno di amplificazione è integrato in tutti i kit PLUS. La scelta appropriata del colorante per il controllo di amplificazione dipende dal tipo di termociclatore su cui la reazione verrà applicata.
