Kits de tests de détection de la contamination microbiologique
Microbiologie
Les aliments contaminés par des micro-organismes tels que des bactéries, des levures, des moisissures ou des virus, peuvent représenter un risque pour le consommateur. En plus de la détection des micro-organismes pathogènes, il est important de surveiller les organismes responsables de l’altération afin de réduire les pertes pendant la production.
Les micro-organismes sont des corpuscules de taille microscopique, généralement composés d’une seule cellule, bien que certains puissent être des organismes pluricellulaires. On les trouve partout dans l’environnement naturel (eau, sol, air, etc.). Les micro-organismes peuvent être naturellement présents dans ou à la surface des aliments (dans les légumes frais, par exemple) ou les contaminer pendant le processus de fabrication des produits alimentaires (hygiène insuffisante lors du traitement de la viande, par exemple).
Dans l’industrie alimentaire, on peut distinguer différents types de contamination microbiologique :
Dans les denrées alimentaires, les micro-organismes pathogènes peuvent ne pas être autorisés du tout ou ne doivent pas dépasser un nombre donné de cellules par gramme d’aliment. Ces limites diffèrent selon chaque matrice alimentaire. (Pour plus d’informations, visitez le site Internet de DGHM). Si les limites sont dépassées, les conséquences pour la santé des consommateurs peuvent être considérables. Par conséquent, une tolérance zéro pour 25 g d’échantillon alimentaire est prescrite pour la plupart des agents pathogènes courants (Salmonella, Campylobacter, E. coli, etc.).
En règle générale, on peut distinguer deux types d’analyses microbiologiques : analyse des aliments et surveillance de l’hygiène / contrôle de la propreté.
Détection de micro-organismes dans l’environnement de production
Dans l’environnement de production, les tests microbiologiques de contrôle de la propreté et de l’hygiène permettent de détecter la présence de résidus alimentaires sur les surfaces insuffisamment nettoyées. Ces résidus de nutriments sont un milieu de croissance idéal pour les micro-organismes.
Le comptage des colonies dans les aliments ou sur les surfaces peut être effectué à l’aide de plaques Compact Dry prêtes à l’emploi. Il existe des plaques Compact Dry pour 13 paramètres d’analyse différents. La méthode de base des examens effectués à l’aide de Compact Dry consiste à cultiver les micro-organismes sur des milieux nutritifs standards. Le principe du test repose sur l’utilisation de substrats chromogènes spécifiques qui sont transformés en produits colorés par le métabolisme des micro-organismes. En plus des échantillons d’aliments dilués, Compact Dry est également adapté aux échantillons prélevés sur des surfaces ou aux échantillons liquides filtrés et incubés.
Les résidus alimentaires prélevés sur des surfaces peuvent être détectés à l’aide de tests de protéines (RIDA®CHECK) ou en mesurant l’ATP (LuciPac™Pen). Le changement de couleur du réactif de détection colorimétrique indiquant la présence de protéines permet de déterminer plus ou moins le stade de contamination. La quantité relative d’ATP (adénosine triphosphate) ou d’AMP (adénosine monophosphate) peut toutefois être mesurée respectivement de façon très précise. En présence d’ATP ou d’AMP, il se produit une réaction de bioluminescence spécifique qui est enregistrée à l’aide d’un luminomètre spécial (Lumitester™ PD-20). Les deux analyses permettent un contrôle cohérent de l’efficacité des procédures de nettoyage. Ces contrôles assurent le maintien de l’hygiène industrielle. Les surfaces bénéficiant d’un niveau d’hygiène parfait réduisent le risque de contamination des produits.

Détection de micro-organismes dans les aliments
Les résidus toxiques des bactéries présentes dans les échantillons d’aliments et de boissons peuvent être analysés à l’aide de kits d’analyse des toxines bactériennes. Pour l’analyse microbienne des aliments, les formats les plus courants sont les tests ELISA, les tests de PCR en temps réel, les plaques nutritives et les plaques de gélose.
Des méthodes immunologiques (ELISA) sont par ailleurs disponibles pour détecter les bactéries pathogènes. Les tests RIDASCREEN® sont basés sur la technologie de type sandwich : l’antigène (protéine bactérienne de surface) est capturé dans le puits d’une microplaque et détecté par l’ajout d’un second anticorps marqué à l’aide d’une enzyme de détection spéciale. La détection immunologique nécessite une procédure de pré-enrichissement appropriée qui dépend du type de bactérie pathogène.
La détection des toxines bactériennes peut être effectuée directement à partir d’échantillons d’aliments (RIDASCREEN® SET Total et RIDASCREEN® SET A,B,C,D,E) ou par détection indirecte de la toxine produisant des bactéries pathogènes (RIDASCREEN® Verotoxin) après un pré-enrichissement approprié.
L’identification des bactéries pathogènes et des virus peut être réalisée à l’aide de systèmes biologiques moléculaires tels que les kits hautement spécifiques de PCR en temps réel de la gamme SureFast® PATHOGEN. Le système modulaire inclut la préparation optimisée de l’ADN à l’aide de colonnes de filtrage centrifuge et de la technique habituelle de sonde d’hydrolyse TaqMan associée à des colorants FAM pour la détection. Il est utilisable avec tous les thermocycleurs courants de PCR en temps réel. Le contrôle interne d’amplification est intégré dans tous les kits PLUS. Le choix du colorant de contrôle d’amplification approprié dépend du type de thermocycleur utilisé pour la réaction.
