Real-time PCR

La PCR (acronimo di reazione a catena della polimerasi) è un importante metodo di analisi per gli alimenti e per la diagnostica clinica.

Un test di real-time PCR duplica e analizza specifiche sequenze di DNA. Nell’analisi degli alimenti i test di PCR trovano impiego in molte applicazioni, come la rilevazione dei microorganismi patogeni, l’identificazione degli allergeni, la rilevazione degli organismi geneticamente modificati (OGM) o l’identificazione delle specie animali. I test di PCR offrono molti vantaggi: sono altamente specifici, sensibili, resistenti, rapidi e affidabili. Inoltre, possono essere automatizzati.

Come funziona la real-time PCR?

Le prime fasi di tutte le procedure analitiche sono il campionamento, che deve essere rappresentativo, e l’omogeneizzazione. Dopo la lisi delle cellule organiche dell’alimento, il DNA viene preparato utilizzando kit di preparazione del DNA o dell’RNA specifici per i batteri o i virus in esame.

I kit SureFood® PREP Basic e SureFood® Prep Advanced sono utilizzati per alimenti solidi e mangimi di origine vegetale o animale. Per le bevande sono utilizzati specifici kit di preparazione del DNA a seconda della matrice, mentre la nuova linea QuickGEN consente uno screening veloce e preciso.

Il sistema modulare di preparazione e rilevazione del DNA può essere utilizzato in modo indipendente. L’amplificazione del DNA bersaglio richiede l’uso, in aggiunta ai kit, di un termociclatore a due (FAM/HEX) o quattro (FAM/HEX/ROX/Cy5) canali di rilevazione.

Campi di applicazione della PCR nell’analisi degli alimenti

Le allergie alimentari possono scatenare reazioni immunologiche anche gravi. Ai fini di tutelare i consumatori, i prodotti alimentari devono essere sottoposti a test rigorosi per la rilevazione di sostanze allergeniche. Offriamo una vasta gamma di kit PCR per l’identificazione di allergeni quali soia, nocciole, arachidi, glutine, sedano, sesamo, senape, crostacei e prodotti ittici. Offriamo inoltre saggi multipli per la rilevazione simultanea di più allergeni (arachidi/nocciole/noci e sedano/soia/senape).

L’identificazione delle specie animali è una questione di grande interesse: i prodotti a base di carne possono essere contraffatti grazie a una errata dichiarazione degli ingredienti, come è successo con lo scandalo della carne di cavallo. Bisogna inoltre garantire l’osservanza di eventuali norme religiose (carni halal e kosher). La nostra linea di prodotti SureFood® Animal ID offre sistemi analitici sensibili per l’identificazione di carne suina, bovina ed equina, oltre ad asino, pollo, tacchino e altre specie. La linea di prodotti SureFood® Fish ID comprende una vasta gamma di kit PCR per l’identificazione delle specie ittiche.

Durante il processo di produzione delle bevande è fondamentale essere in grado di rilevare eventuali istanze di degradazione. La linea GEN-IAL® include una vasta gamma di kit per la rilevazione, nel corso della produzione di vino e birra, di organismi di degradazione quali Lactobacillus, Pediococcus, Megasphaera, Pectinatus, Dekkera bruxellensis ed Oenococcus oeni. I kit possono essere utilizzati durante il processo produttivo o per esaminare i prodotti finiti.

Negli ultimi anni la produzione di organismi geneticamente modificati ha avuto una notevole impennata in tutto il mondo. La presenza di OGM nei prodotti alimentari è strettamente regolamentata, e c’è grande richiesta di metodi analitici sensibili in grado di rilevarli. La nostra linea di prodotti SureFood® GMO offre kit per la rilevazione qualitativa e quantitativa di OGM quali mais geneticamente modificato, riso LibertyLink e soia Roundup ready.

Uno dei campi principali di applicazione delle tecnologie PCR nell’analisi alimentare è il rilevamento di eventuali contaminazioni microbiologiche. La nostra linea di prodotti SureFast® è in grado di identificare una vasta gamma di batteri patogeni (Salmonella, Listeria, Legionella, E. coli, Staphylococcus aureus, Campylobacter), virus (norovirus, virus dell’epatite e dell’influenza), lieviti e muffe.
